注意事項:
1、產品驗收:
① 血清內外包裝均應完好,無破損、裂縫、溢滲等現象;
② 血清到貨時應為冷凍或冰水混合狀態,不應完全融化;
③ 若出現上述現象,請拍照取證后及時與我們聯系,以便及時進行更換。
2、血清的解凍:
① 請在2~8℃環境中解凍,不宜在較高溫度下進行解凍;解凍溫度較高會導致血清渾濁,沉淀增加,品質下降;
② 血清解凍的過程中請不時搖勻(小心勿造成氣泡),使血清成分和溫度均勻,從而減少沉淀的產生;
③ 血清解凍后應盡快用完,盡量避免反復凍融;
④ 血清不宜在室溫中長時間放置,使用后盡快放2~8℃中;
⑤ 高溫解凍、劇烈搖晃、反復凍融、放置時間過長等,均會導致血清品質的降低。
3、運輸及儲存條件
運輸條件:冷凍、密封、避光;
儲存條件:貯藏溫度≤-15℃, 可長時間保存;2~8℃存放時,請勿超過一個月。
FAQs:
Q1:如何解凍血清才不會使產品質量受損?
A1:① 將血清從低溫冰箱取出后,先于2~8℃冰箱放置12-24小時左右使之部分溶解,然后在室溫下使之全溶;但必須注意的是,溶解過程中必須規則地搖晃均勻。
② 切勿將剛剛從-20℃冰箱里拿出來的血清直接放在水浴中,無論室溫的水或者37℃的水;因為在水浴中血清迅速融化,過大的溫差變化(-20℃→37℃,溫差為57℃)極其容易造成血清析出沉淀。
③ 血清應該置于-15℃以下保存;若一次無法用完一瓶,應該無菌分裝,再冷凍保存,避免反復凍融。
Q2:血清中可能出現的沉淀物是什么?
A2:① 纖維蛋白,它是經常出現的較大的沉淀物,可以達到1-2mm,可以用肉眼觀察到。
② 磷酸鈣,它也是常見的一種沉淀物,通常會使血清出現渾濁,并且在37℃培養的時候會增加。這種沉淀物在倒置顯微鏡下觀察像小黑點,這些小黑點由于布朗運動看上去可以活動,因此經常被誤認為是微生物污染。
③ 膽固醇、脂肪酸酯以及一些蛋白質,它們也是血清中出現沉淀物的常見原因。關于細胞生長,我們的試驗以及經驗表明沉淀物不會影響細胞培養,我們的客戶以及其它血清生產商也證明了這一點。
Q3:血清解凍后發現有絮狀沉淀物出現,該如何處理?
A3:若您欲去除這些絮狀沉淀物,可以將血清分裝至無菌離心管內,以400-600g離心5min,上清液即可加入培養基內一起培養。 我們不建議您以過濾的方法去除這些絮狀沉淀物,一方面它可能會阻塞您的過濾膜;另一方面,過濾血清這種行為可能會導致血清中部分營養成分的流失。
Q4:如何避免沉淀物的產生?
A4:溫度過高、時間過久、搖晃不均勻等,都會造成沉淀增多;我們建議,如非必要,無須對血清進行滅活;若必須熱滅活,應嚴格遵守56℃、30min 的原則,并隨時搖晃均勻。
Q5:保存血清最好的方法是什么?
A5:需長期保存的血清必須儲存于-15℃或更低溫冰箱中;研究表明,儲存在-80℃下的血清在性能方面沒有任何變化,但解凍時巨大的溫差會導致更多沉淀的產生,故不建議-80℃儲存。血清在2~8℃冰箱存放時間切勿超過1個月;若一次無法用完1瓶,建議分裝后保存,避免反復凍融。另外,血清結冰體積會增加約10%,分裝時請預留一定的體積空間。
